Adenosine receptor (AdoR) inhibitors decreased cordycepin-promoted acrosomal reaction (AR) and hyperactivation in human sperm. (A) Human sperm was stained using fluorescein isothiocyanate-conjugated Pisum sativum agglutinin (FITC-PSA), which can distinguish the AR and acrosome integrity (AI) easily. Sperm were treated with cordycepin and AdoR inhibitors separately or simultaneously. Inhibitors to the A1 (DPCPX), A2a (DMPX), A2b (MRS1754), and A3 (MRS1191) AdoRs were used. The PKA inhibitor H-89 was used as the positive control. (B) Statistical analysis of AR ratio. Data are presented as means ± standard deviation (SD; n = 3). *P < 0.05 (compared with 500 µM cordycepin group). (C) The effects of AdoR inhibitors on hyperactivation. The PKA inhibitor H-89 was used as the positive control. Data are presented as means ± SD (n = 5). *P < 0.05 (compared with 500 µM cordycepin group)
